The HotStart tubes automate the HotStart PCR process by using a pre-positioned wax barrier to separate key reagents until the annealing temperature is reached, there by decrease the occurrence of misprimes, primer-dimers, and premature annealing. The unique combination of thin-walled tube and pre-positioned wax bead optimize the hot start reaction and eliminates the need for messy oil overlays.

HotStart tube can also be used for the storage of commonly used primer sets and PCR product for months for later use or analysis. Simply create a master mix, aliquote into the HotStart tubes, melt the wax bead, and freeze! This is a great way to save time and ensure test-to-test consistency when regularly running several of the same reaction types using identical primer sets.

HotStart’s unique design can also facilitate a, one-tube RT-PCR procedure by using a thermostable reverse transcription enzyme. Simply add your revere transcription reagents to the lower layer and the PCR reagents to the upper layer. The first denotation cycle of PCR will melt the wax, denature any side reagents from the RT reaction and begin the amplification of the target cDNA.
